Design Notes
Due to the presence of multiple high CG content regions in this particular part, it posed a challenge for direct synthesis by IDT. Despite optimization efforts, we still had to divide the sequence into three separate gBlock fragments for ordering. This approach allowed us to overcome the difficulties associated with the high CG content regions and proceed with the assembly process.

The original tdTomato sequence was obtained from NCBI. Then, the sequence was codon optimized by IDT and gBlock was synthesized from IDT.
