The primer was designed to perform site-directed mutation which was defined as a forward primer. It can mutate base sequence which is on node 59 and coding the 79th amino acid from glutamine to aspartic acid. According to Codon preference in E.coli, AAT has the highest possibility coding aspartic acid.
