Design Notes

All sequences are comaptible with RFC1000, the iGEM type IIS assembly standard. Previous teams (2012 Darmstadt) have submitted basic CDS parts used in this system, but not all parts were RFC1000 compatible. In addition, because we ordered this composite part for assembly, we codon optimized it as well to avoid any complexities when ordering the gene fragments from Twist Bioscience.

A TAA stop codon is added after each CDS to give a double stop. This page records the second stop codon as a "scar".

The CDSs come from Comamonas testosteroni, and the promoter, RBS, and terminator comes from the 2023 iGEM distribtution kit.
