Design Notes
As one of the most important synthetic biology components in transcriptional regulation, promoters play an irreplaceable role in metabolic engineering. For the industrial microorganism Corynebacterium glutamicum, the construction of a promoter with a strong promoter is essential for the production of the target product lycopene. The CRISPR-MAD7 nuclease was ligated to the Ptrc promoter, and then IPTG was added under appropriate conditions to achieve the regulation of CRISPR-MAD7 expression. So that the Ptrc promoter can better regulate the expression of CRISPR-MAD7, so that is can play a greater role in Corynebacterium glutamicum. Up to now, IPTG-induced Ptrc is still applied to exogenous inducible promoter in Corynebacterium glutamicum.
