Design Notes

When incorporating the riboswitch in constructs like our composite part BBa_K4654018, we made sure to include the spacer sequences up- and downstream of the riboswitch to preserve the natural environment of the riboswitch, because they influence the folding of the riboswitch.

The riboswitch can be found in certain bacteria, the sequence we are using was created by White et. al. by creating a consenus sequence from different sequences of the riboswtich family1.
