Fig1. The HindIII cutting site (AAGCTT) is on the stop codon, and there is only one BamHI cutting site on the plasmid. Therefore, if we mutate the TAA stop codon correctly, the result will be one band (3238bp) when we digested with BamHI and HindIII.
