Design Notes

Sheffield 2022 designed this part for testing the function of their MutaT7-based directed evolution system ‘rEvolver’. Since this system was designed for use in V. natriegens, BBa_K4451015 has been codon-optimised accordingly. This part is used in the composite ‘MutaT7 Test Cassette’ BBa_K4451022, where streptomycin resistance above that of the spontaneous mutation rate acts as a reporter for adenosine deaminase activity (caused by a A → G transition mutation; see BBa_K4451022 for more information).
