Design Notes
The genome of Azotobacter vinelandii for example shows a high GC content, which can be a problem in the synthesis. Future teams should consider finding a matching polymerase since the commonly used T7 promoter is not compatible. Since the fragment is quite long a two part amplification would be suitable.
