Histidinol Dehydrogenase
Histidinol dehydrogenase is an enzyme needed for the biosynthesis of the amino acid histidine. It specifically catalyzes the reaction L-histidinol + H2O+ 2 NAD+ L-histidine + 2 NADH. Because histidine is necessary for the survival of the cell, a system for selectivity can be designed by engineering an auxotrophic mutant strain with a deficient copy of the his4 gene. Synthetic biology operations are then possible by constructing a plasmid providing both the functional his4 and the part of interest. In histine absent media, cells will only grow following successful transformation
This part occurs in nature with three unwanted restriction sites(XbaI sites @ 1941 and 2474 as well as a BamH1 site @ 275). The part displayed here shows the intended finalized part after the site directed mutagenesis correction of these sites.
