Design Notes

The whole sequence of Kozak sequence is CCGGGTGGTCAGTGCCACCATGG. As the sequence includes a start codon, this sequence should be cut at C|ATGG at golden gate assembly. Thus, the former scar of CDS by SapI should be ATGG.
