Applications of BBa_K4296000

Our team (UBC Vancouver iGEM 2022) used this part to design a genetic IF circuit where the condition was temperatures above 37C. Only at high temperatures does the wheat transcription factor TaHsfA2b bind to the promoter TaHsp70d, activating it. This promoter can be placed upstream of genes you would like to express only at high temperatures. At lower temperatures there is low to none expression of downstream genes. This promoter requires the wheat transcription factor TaHsfA2b so for our project we used wheat protoplasts to test the expression of our gene constructs. Wheat protoplasts are plant cells with their cell wall removes, making them easier to transform.
