Design Notes
1. The GAL4 Promoter from K.marxianus is assembled into the secretory protein expression vector via goldengate assembly, with two BsaI cleavage sites and two unique overhangs.
2. To construct a K.marxianus secretory expression vector, the plasmid backbone, pLAC4 promoter, MFalpha, mTurquiose and tTDH1 terminator are firstly assembled as the vector for later assembly operations.
