Design Notes
First, we need a plasmid with two operons, because we need to insert two different promoters into it; The temperature sensitive regulatory element cI857 is placed after the ordinary promoter, and the inducible promoter R induced by cI857 is placed after pst to achieve the expression regulated by temperature. Secondly, we need to achieve the co expression of the two genes after the same promoter. For this, we specially consulted the research group of Chen Guoqiang from Tsinghua University and got a solution. Finally, we mainly used mCherry to verify that cI857 is temperature regulated, so we recombined the fluorescent protein homologously to the position of pst.
