PcysJ is regulator controlled by threonine concentration, and its downstream gene expression activity increases with the increase of threonine concentration.
MazF is a non-specific ribonuclease (toxin) in E. coli and MazE is an antitoxin that combines it.
The system was used to construct a suicide switch in response to threonine concentration. When the concentration of threonine was high, MazF and MazE were co-expressed, and the two neutralized each other to ensure that the bacteria would not die. However, when the threonine concentration decreased, the expressions of MazE and MazF decreased simultaneously, but because the stability of the antitoxin MazE was lower than that of MazF, more free MazF in the bacteria would kill the bacteria.
