Design Notes
I need to confirm whether I need a repetitive domain for surface protein to function and whether it is added at N or C terminus. There are many surface display systems in L.lactis, and the LysM system is tested in displaying anti-TNF-alpha nanobodies. As Visweswaran et al. recently reviewed, the peptidoglycan binding properties have been extensively utilized for surface display. Therefore, using a repetitive LysM system is likely to be successful.
Source
The sequences come from L.lactis NZ9000 genomic DNA.
