OR gate
OR gate is a type of Boolean logic gate that can be used to activate downstream gene expression in response to an upstream counterpart promoter in the presence of any or all markers.
Usage and Biology
The OR gate is constructed with a p7 promoter and a terminator ( BBa_B0015 ). We added all cis Bxb1 integrase and Tp901 integrase sites on both sides of the terminator to form the so-called OR gate. Finally, we introduced RiboJ and BCD2 sequences to ensure smooth transcription with the OR gate on.
The OR gate can be assembled to any biobrick containing an RBS site. You can use the OR gate to respond to multiple promoters upstream, in the presence of any or all of the corresponding markers. To demonstrate its efficacy, we designed the RFP protein downstream of it to test it ( BBa_K4156124 ).
Meanwhile, we plan to apply the OR gate on the expression of HlyE ( BBa_K4156125 ). Together with the use of upstream regulatory sequences, we can control the expression of HlyE only when needed, while avoiding any undesired production and harm.
