To express our target proteins, the phospholipase A2 (BBa_K4091024, the part that this page describes) and the gamma phospholipase inhibitor (BBa_K4091069), we use the T7 promoter in our circuit in order to induce the protein expression by adding IPTG (Isopropyl β-d-1-thiogalactopyranoside) in the media. We did so to perform the inhibitory assay to quantify how the kinetics of this enzyme truly works, however, we could not finish our experiments due to the fact that our distribution kit did not arrive. We know that the inhibitory mechanism of this particular interaction follows a competitive inhibitory kinetic, so we could make a mathematical model to predict some results. Once we could get our data, we would have the equation that describes the behavior in which the substrate is consumed through time. We analyzed the results via SDS-PAGE to confirm whether or not if we could express our proteins.
