Design Notes
This is the positive control of the APOB gene that will be used within a Flappase genetic testing system.
An illegal SpeI restriction site was found to be present in the ERV2-1 transposon insert. To alter this to make the part compatible with the RCF10 assembly, the sequence ACTAGT within the insert has been changed to GCTAGT. This eliminates the illegal restriction site.
Source
The source of the APOB-100 sequence is the genomic sequence from Bos taurus, apolipoprotein B genome.
References
Through the assistance provided by Team Rochester, we were able to acquire the ERV2-1 sequence from their subscription to Repbase. Repbase is a database consisting of repetitive DNA elements.
