The cellulase plasmid expresses the cellulase with Type1 dockerin that can be secreted to the outside of the cell, and the cellulase is connected to the dockerin through a flexible linker (GGGGS)3. The Type1 dockerin derived from Clostridium cellulovorans can be combined with the Type1 cohesion on the skeleton to anchor the cellulase on the skeleton. In addition, there should be a tag on the cellulase plasmid. The short peptide expressed by the tag can be combined with the primary antibody and the secondary antibody with fluorescent dye to verify the binding of the cellulase to the backbone in the follow-up.
