Design Notes

The native CipA Scaffoldin is from Acetivibrio thermocellus (NCBI ACCESION is WP_020458017) and it has night cohesion, one CBM, and one Type II dockerin. Based on the natural CipA protein, we changed Cohesion1 and Cohesion3 to Ccscohesion, kept the Cohesion4 as Ctcohesion, kept the CBM3 domain, and deleted Cohesion2 and Cohesion5-9. Finally, we designed a scaffoldin protein by ourselves. It has two Ccscohesion, one CBM3, one Ctcohesion, and one Type II dockerin. The Ccscohesion can bind with Ccsdockerin. Similarly, the Ctcohesion can bind with Ctdockerin. We can connect dockerin with other enzymes,so that the enzymes with dockerin can bind to the scaffoldin protein and work synergistically.
