Applications of BBa_K3747141

Nos is a nitrous oxide reductase, which is required for denitrification [1]. Nos is the last enzyme in the denitrification pathway and reduces nitrous oxide (N2O) into dinitrogen gas (N2). We expressed this construct (BBa_K3747140)in Pseudomonas putida EM42 together with a plasmid containing possible electron donors for Nos to establish denitrification in this organism.

As the nos construct contains a rhamnose inducible promoter, the optimal rhamnose concentration (0.23 mM) by performing a growth experiment in which the OD600 was measured continuously over time (Figure 1).

Figure 1. Induction of nos operon in P. putida EM42 with different rhamnose concentrations. P. putida EM42 containing pSEVA22-NosRZDFYLTatA and pSEVA83- ApbE-blue copper protein 1 – blue copper protein 2 – c550 (Nos). As control, P. putida EM42 containing pSEVAb22 without cargo was used (Control). Three biological replicates were used for the control and two biological replicates for Nos.
