Design Notes
Users can change the 15 bases at the 3 'end according to their own needs, and replace them with the sequences that their machines want to recognize. The length of 5'oligoT of DW_lineL can be appropriate, but it should not be too long or too short. The length of 3 'recognition sequence can also be designed according to the demand, but the data show that the total length of recognition sequence of lineL and lineW should not exceed 40 nt and should not be shorter than 20 nt. On average, the sequence of lineL recognition region should be 10 to 20 nt.
Source
The 3' end 15 nts sequence of DW_lineL is comes from a detection site of N gene of SARS-CoV-2.
