Our targeted gene is SLCO1B1 which is located on chromosome 12 (Chromosome 12: 21,131,194-21,239,796 forward strand). The gene fragment that was amplified contains the sixth and the seventh exon of the gene. The studied variant is located in the sixth exon.
Usage and Biology
This gene fragment was amplified in order to detect the targeted variant rs4149056. If this fragment was produced using the Forward primer for the wild type (Part:BBa_K3464000) and the Reverse (Part:BBa_K3464002), indicates the existence of the T allele. Respectively, if the fragment was produced using the Forward primer for the alternative allele (Part:BBa_K3464001) and the Reverse (Part:BBa_K3464002), indicates the existence of the C allele. The fragment’s length is 535bp (Part:BBa_K3464003).
We had to take in consideration the stability of the secondary structure DNA PCR products, mainly by using thermodynamic methods via the The DINAMelt Web Server "DINAMelt".
