AIP generator (agrB and agrD), produce AIP

The agrB gene encodes a peptidase used to process the signaling pro-peptide. The agrD gene encodes signal pro-peptides.

Usage and Biology

The expression of virulence factors in Staphylococcus aureus is strictly controlled by an auto-induced quorum sensing system called the auxiliary gene Regulator (Agr) system. The agrD gene encodes the signaling propeptide, and the agrB gene encodes the peptidase used to process the signaling propeptide. AgrD mature peptide is the signal peptide of Agr system, also known as self-induced peptide (AIP) molecule. AgrC and AgrA constitute a two-part system. AgrC is the receptor for AIP. Upon binding to AIP, AgrC dimerizes and phosphorylates each other to activate AgrC proteins. AgrC can phosphorylate AgrA protein. The phosphorylated AgrA protein dimerizes and binds to the P2 promoter of the agrBDCA operon to form a positive feedback loop. The activity of AgrA dimers will also combine with P3 promoter, to activate the non-coding RNA, the RNA III expression. RNA III can affect the virulence factor and adhesion factor of the translation process.

We use this part to produce AIP. AIP generator (agrB and agrD) and AIP sensing device (agrA and agrC) were introduced into different cells to achieve the effect of acting on each other through AIP, and the corresponding purpose was achieved by connecting the target genes downstream of P2 promoter.
