agrA and agrC, an AIP sensing element

The sensing element of AIP.


Usage and Biology

AgrC and AgrA constitute a two-part system. AgrC is the receptor for AIP. Upon binding to AIP, AgrC dimerizes and phosphorylates each other to activate AgrC proteins. AgrC can phosphorylate AgrA protein. The phosphorylated AgrA protein dimerizes and binds to the P2 promoter of the agrBDCA operon to form a positive feedback loop. The activity of AgrA dimers will also combine with P3 promoter, to activate the non-coding RNA, the RNA III expression. RNA III can affect the virulence factor and adhesion factor of the translation process.

In our project, We realize the induction to AIP by constitutive expression of agrA and agrC. The effectiveness of the system was validated by attaching fluorescent proteins downstream of P2 BBa_K3447011.
