Design Notes

This RNA aptamer has not been tested in the lab by our team, however the Edinburgh iGEM 2020 team tested the BBa_K3380150 RNA aptamer in the BBa_K3380500 construct and BBa_K3380600. Similar design can be used for the Broccoli RNA aptamer.

All the sequences for the individual parts were acquired. The individual part were designed with 4bp scars to allow swapping of the parts to test different constructs. The individual parts were phosphorylated, annealed and ligated together using the T4 ligase.

Filonov et al. 2014 used directed evolution to obtain the 49 nucleotides Broccoli fluorescent RNA aptamer.
