This part is one of the targets of our Moloney Murine Leukemia Reverse Transcriptase (MMLV RT https://parts.igem.org/Part:BBa_K3257042). R region, polypurine tract, primer binding site and U5 region help with the reverse transcriptase binding to the target and initiating rounds of reverse transcription while lox sites enable Cre recombinase (https://parts.igem.org/Part:BBa_K3257044) to recognize and finish homologous recombination. The EGFP (https://parts.igem.org/Part:BBa_E0040) we use is a reporter protein that has been characterized by a lot of former iGEM teams.
