DNA program 12346

This part contains a DNA program - a specifically designed nucleotide sequence containing 5 zinc finger binding sites. The program was designed in such a way, that chimeric enzymes (fused with zinc finger proteins) for synthesis of violacein bind to the sequence in the order required to produce the final product. The part contains binding sites for zinc finger proteins in the following order: Jazz - Blues - Zif268 - PBSII - Gli1.

We have bound four out of five chimeric enzymes (Blues_VioA, Zif268_VioB, PBSII_VioE and HivC_VioD), encoded by Part:BBa_K323135 and Part:BBa_K323132, to the program sequence and have shown that the order of the enzymes bound on the DNA program determines the final product by directing metabolic flow.
