Design Notes
The chimeric GPA1-Gαs is made to be compatible with the human GPCRs (in our case GPER, Hu-LHCGR & XLHCGR). The chimera acts as an adaptor molecule to mediate the conformation change in the receptor to the yeast endogenous beta subunit Ste4p, and gamma subunit Ste18p. It has been shown that changing the last 5 amino acids in the C-terminus of the coding sequence of GPA1 is enough to make the chimera work efficiently and that is the same approach we followed.
