Design Notes
This part is consisted of Vector pTRE, L7Ae and miR592-target. There is a CMV promoter on pTRE that can transcribe mRNAs with L7Ae and miR592-target. The translation of this mRNA is controlled by the amount of intracellular free miRNA592. When miRNA692 exists, the translation of this mRNA is suppressed. The mRNA gene product is L7Ae, which can combine with Kturn structure on mRNA to suppress the translation of that mRNA, acting as a regulatory part.
pTRE-L7Ae-miR592 is assembled into our circuit. When miRNA592 exists in the cell, the translation of L7Ae is suppressed and L7Ae cannot be produced whereas the mRNA with Kturn structure can be translated to express the response part.
