sRBS (30000)

sRBS (30000) is a strong ribosome binding site present upstream of the transcriptional unit. sRBS (30000) has a very strong binding affinity with the ribosomes resulting in enhanced protein expression.

Biology

A Ribosome Binding Site (or RBS) is a short DNA sequence present upstream of the transcriptional unit. In prokaryotic systems, it is easily identified by the presence of a Shine-Dalgarno like sequence which pairs with the anti-Shine-Dalgarno like sequence of the 3' end of 16S rRNA. An RBS is crucial for the process of translation as it dictates the binding of the ribosome (the translational machinery) to the mRNA transcript encoding the protein of interest. The effectiveness (or strength) of the RBS sequence depends on the base-pairing potential of the RBS to the 16S rRNA along with the spacing between the sequence and the start codon. sRBS (30000) happens to be a strong ribosome binding site enhancing translational efficiency.

Usage

The sRBS sequence can be used upstream of any gene whose product is desired to be enhanced. This part is used to maximize the translational efficiency of the CcaS protein in the composite gene constructs.
