Design Notes

It is needed to be complementary to other oligo from parts BBa_K3087000, BBa_K3087002 and BBa_K3087003 to form the Lactobacillus targeting nano-structure.

The design of the Lactobacillus targeting nano-structure plasmid are as follow:

The biobricks contained in our plasmid are illustrated in the above graph. Four basic parts we designed, including BBa_K3087000, BBa_K3087001,BBa_K3087002 and BBa_K3087003, are the DNAs synthesised to form our Lactobacillus targeting nano-structure (LTN).

The promoter used in these four basic parts is J23100 and the terminator used is B0054. HIV Terminal Binding Site (HTBS) used in each plasmid encode the end of our desired gene. The binding of HIV reverse transcriptase (HIVRT) would then reversely transcribe our target DNA.

The validation of plasmid is shown above. O1, O2, O3 and O4 represent the part BBa_K3087000, BBa_K3087001, BBa_K3087002 and BBa_K3087003 respectively.
