LOV2: NLS-VP16-EL222
This part codes for LOV2, a light-inducible transcription factor. It is made up of an NLS fused to VP16 and an EL222 region. When LOV2 is energized by blue (450-490 nm) light, it undergoes a conformational change that exposes a helix-turn-helix (HTH) DNA binding domain. The VP16 sequence of LOV2 also acts as a transcription factor, inducing the transcription of a downstream sequence. For example, an organism expressing both BBa_K2805002 and BBa_K2805003 will produce mRuby, a red fluorescent protein, when under a blue light. This sequence is optimized for use in S. cerevisiae.
Additionally, organisms expressing this plasmid should be kept in the dark. White light will also activate LOV2.
