Based on sequencing after using this part, it is possible that the first base of the biobrick suffix in the vector may have mutated from T to A. This doesn't impact the SpeI site, so should be a harmless issue.

We ended up not characterizing our constructs (K618111 and K618112) as thoroughly as Bristol, but observationally, uninduced cells with the pBAD promoter grew better than uninduced cells carrying the pLAC promoter (K618211 and K618212). Thus, this promoter may be a good choice when you desire very low basal expression. For membrane fusions where toxicity is a concern, this is an asset.
