B7-1 Murine Trans Membrane Domain
The b7-1 cytoplasmic domain enhances transport from the endoplasmic reticulum and allows accumulation of chimeric proteins on the plasma membrane. It has the same function as PDGFR (the human platelet-derived growth factor receptor), anchoring heterologous proteins on cells' membrane. We used scFv (single chain fragment variable) fused to B7- 1 TM (trans membrane) domain, which according to the literature has increased the expression of scFv on baby hamster kidney cells, approximately 7 fold greater than scFv-PDGFR.

Figure 1: Membrane-anchored single-chain antibodies. A scFv is composed of the immunoglobulin heavy and light chain variable regions linked together with a flexible peptide (normally 15 amino acids in length). For expression of a membrane-anchored scFv, a signal peptide (SP) directs the nascent chain to the lumen of the endoplasmic reticulum whereas the TM anchors the antibody in the plasma membrane.
