nrfHA Composite Part
This gene encodes both Cytochrome c552 (nrfA) and Cytochrome c small subunit (nrfH). The part includes the constitutive promoter J23100, the medium strength ribosome binding site B0032 before each coding sequence, and the double terminator B0015.
This part was designed to convert nitrite to ammonia within bovine rumina. The purpose is to increase cattles’ tolerance of nitrate/nitrite in their diet(s). Overexpression of this gene is also proposed to help redirect electron flow away from methanogens, thus reducing methane production from cattle. The coding sequences of nrfHA come from Selenomonas ruminantium subsp. lactilytica TAM6421 registered on Genbank. S. ruminantium is a gram-negative bacteria native to bovine rumina. The composite part was fully sequenced and showed no mutations
