This part is an expression vector that includes the coding sequence for rtTA3, a DOX-inducible transcription factor that regulates the Tet Response Element (TRE) promoter. This sequence lies downstream of the human elongation factor 1a (hEF1a) promoter, which is a low level constitutive promoter. Our team used other parts that make use of the TRE promoter so DOX induction in addition to this construct could be used to control the activity of the promoter. More specifically, the TRE promoter was used upstream of our RBP gene sequences to control and reporter constructs, and changed the amount of produced depending on the amount of DOX used applied to the system.
