CEN/ARS

Compatibility of a circularized E. coli plasmid with the yeast DNA replication machinery can be gained by introducing certain DNA sequences called autonomous replication sequences (ARS). Circularized plasmids that can be replicated and propagated in two different organisms are called shuttle vectors because the genetic material they carry can be transferred from one to the other. Several different ARS are used frequently in the yeast community, but none are in the standard BioBrick format.

Replication and propagation can also be enabled by sequences that allows linearized DNA to be integrated at targeted sites in the yeast genome. Plasmids that allow integration into the genome are called targeting vectors. Several are used frequently in the yeast community, but none are in the standard BioBrick format.


Composed of yeast centromere (CEN)and an autonomously replicating sequence (ARS); This part can be combined with BBa_K2327002 and BBa_K2327003 to create yeast shuttle vertors with different selection cassettes, allowing propagation of genetic material in both yeast and e.coli.
