unc-60 RFP reporter unc-60 encodes two differents tissus- isoforms of actin depolymerizing factor (ADF/cofilin). These proteins are actin-binding proteins that regulate filament dynamics. Indeed the first isoform, UNC-60A binds G-actin and depolymerizes actin filaments and which function is to inhibit actin polymerization. UNC-60B binds F and G-actin and  influences the rate of actin polymerization with the UNC-60B binding ratio. Two Signal transduction and activation of RNA proteins (STAR proteins) are involved in the specific splicing of this gene. These proteins are ASD-2 (Alternative -Splicing-Defective-2) and SUP-12. ASD-2 and SUP-12 cooperatively bind to CUAAC repeats and UGUGUG stretch in intron 1A of the Pre-messenger RNA in order to switch the UNC-60A type to UNC-60B type in body wall muscle. To study the alternative splicing of this gene and see in vivo if UNC-60A (non muscle isoform) is produced, the unc-60 RFP reporter design by Hidehito Kuroyanagi  can be used to study  in space and time if this isoform is present into the different tissues of Caenorhabditis elegans. Indeed this part contains introns 1A between Exon 1 and Exon 2A in fusion with the RFP protein. These exons are essential in this part for the reason that if both ASD-2 and SUP-12 are present, they can bind intron 1A and no splicing is happening. As a consequence, no RFP signal is present. But if one of these proteins are absent, the alternative splicing is possible and one RFP signal is present.
