Design Notes
When designing this insert, it was initially intended to be used in the pDB.His.X series plasmids and the pET-28 derived plasmid pTHST. The pDB.His.X series plasmids contain the T7 promoter, a 6xHis tag, followed by an expression tag (GST, MBP) and a TEV protease cleavage site. Expression of our insert can be induced by adding IPTG to a liquid culture of E. coli transformed with our construct. Since our construct already contained an expression tag, a promoter, and a RBS in the vector, these were not included in the design of our insert.
