Applications of BBa_K199135

This device was designed as a positive control for our SAT problem solver [http://2009.igem.org/Team:MoWestern_Davidson see 2009 team wiki]. Unfortunately, the three TT appear to bring instability to the DNA. While maintaining the cells, the insert is recombined and the part is very unstable. Using 3 TT should be avoided by others.

We used an optimized LuxI K199125 and it worked perfectly. When plated on LB media, or grown in liquid broth, the JM109 cells turned red as predicted since pLac is a leaky promoter.

(a) Positive feedback loop with wt optimized LuxI (BBa_K199135) plated on LB-Amp. Some regions of plated cells lack any red fluorescence (b) pLpp-RBS-RFP. K199135 cells were initially more similar in colour to pLpp-RBS-RFP.

Solving a 1 SAT problem with suppressor tRNAs.
