Ptet+RBS+LuxI(LVA)+terminator

We designed an economical mechanism that can count the population of invading bacteria at real time with LuxI/LuxR device. The counter function in engineered E.coli can remind us the situation of environment by different color fluorescent proteins. Green fluorescent light means the counter system is turned on and environment is safe; the yellow fluorescent light means the system is going to lose freshness; and red fluorescent light indicates system has bacteria contamination and the environment become dangerous!

This device is for outer bacterial invasion detectionand time delay function of our timer project. LuxI can be produced to synthesis AHL when tet promoter is not inhibited. this is the C device of NCTU-formosa 2009 project.

The device not only can detect invading bacterial but also can sense the amount of our desighed E(by AHL). Coli. By this way, the amount of desighed E coli. Itself can influence the duration of our timer function. So the growth of our desighed E.coli amount would shorten timer duration. This function would work at consumption phase of our project.

The α factor is the amount of AHL(bacterial connection factor) no matter form E. coli itself or invasive bacteria. They bind with luxR and bind to luxR/cⅡp22 Promoter, than the F device produces RFP.
