Applications of BBa_K1711001
After transforming our theophylline-responsive system into yeast, we used a flow cytometer to measure the fluorescence of our system, both in the presence of theophylline and without it. Our initial results from the experiment with the theophylline induced and non-induced conditions showed a systematic difference with all induced trials having a higher mean fluorescence per cell. Yeast transformant 3 showed a greater response than the other transformants.
Figure 1: Fluorescence was systematically higher for samples with theophylline than with no theophylline.
Next, we wanted to find out if fluorescence levels were dosage-dependent. Our subsequent concentration dependence experiment showed an increase in fluorescence with increasing concentrations of theophylline. Here again the transformant 3 showed the best concentration dependence.
Figure 2: As the concentration of theophylline increased, fluorescence reading increased for each of our yeast variants.
