Design Notes
KIG is a fusion protein without scar inside, constructed from Kcoil, VMA and green fluorescent protein. Kcoil is a synthetic coiled-coil domain designed specifically to bind to Ecoil with high affinity and specificity. VMA is a thermosensitive intein which can self-spilt at 18℃. We design it to use cooperatively with BBa_K1189018(2013,Calgary). What's more, we added BamHI and SalI to the flank of the fusion protein, so that users can change the part to plasmids which for protein expression easily.
