micF Promoter, Sensing Antibiotics

Usage and Biology

Description

micF, an antisense ncRNA targeting ompF RNA, main endogenous porin of Escherichia coli, participating in post-transcriptionally regulation of this outer membrane protein F(ompF). micF is also regarded as one of the promoter of soxS-regulated gene.

micF was originally tested by Sahar Melamed and Shimshon Belkin to research the antibiotic resistance of bacteria and features of selected sensing promoters. Sensitivity and specificity of micF was presented, showing its relatively specific signal response when treated with:

  • sulfamethoxazole-255.2;
  • sulfadimethoxine-44.9;
  • colistin-36.9.


The maximal induction of micF strains tested in the course of 10h of exposure.

Test Result

Application

In accordance of micF being responsive to the target compound, we implement micF to sense the existence of sulfamethoxazole and sulfadimethoxine in watersample.

Experimental Result

micF and soxS, these two antibiotic substance responding promoters were extracted from E. coli Top 10(K-12 strain) by conducting polymerase chain reaction on its genome.
