Design Notes
The assembly was performed by GoldenBraid 2.0, a Golden-Gate based assembly system. The barnase was amplified by PCR from Part:BBa_J72173 and a mutation in T105P was introduced. The vector used to perform the assembly was Part:BBa_K1554006, a vector with the BioBricks backbone pSB1A3 and compatible with the Golden Gate-based assembly system GoldenBraid 2.0.
