Applications of BBa_K1408001

For The University of Washington's 2014 iGEM project we utilized Gal4-Vp16 to up-regulate a GFP gene under the Gal1 promoter found in the PyE1 strain of yeast. The bar labeled as "No insert" "Deg0" in figure 1 refers to only Gal4-Vp16 up-regulating GFP expression. Other bars represent Gal4-Vp16 fusion proteins with a degron (an unstable protein domain) inserted in 1 of 4 positions and a protein with its own native stability. Figure one shows that Gal4-VP16 on its own increases the expression of genes under the Gal1 promoter much more than if it is being degraded due to unstable components fused to it.
