Applications of BBa_K136009

MG1655 (envZ-) was transformed with pSB3K3-BBa_K136066 (pBAD-envZ*) and BBa K136009 in BBa_J61002. In J61002, the promoter regulates the expression of mRFP1. The J61002- K136009 was here modified to put the mRFP1 in frame. This was done through modification of the SpeI site by digestion, klenow reaction and ligation.
EnvZ (expressed from the pBAD) naturally represses flhDC through the activation of OmpR. FlhDC is the master regulator of the feed forward loop leading to the expression of the flagella class 2 genes (fliL, flgA, flhB...). We measure here their activity with envZ expressed 0.2% arabinose or repressed 1% glucose.
