Applications of BBa_K136006

MG1655 (envZ-) was transformed with pSB3K3-BBa_K136066 (pBAD-envZ*) and BBa K136006 (pflgA) in BBa_J61002. In J61002, the promoter regulates the expression of mRFP1.
EnvZ (expressed from the pBAD) naturally represses flhDC through the activation of OmpR. FlhDC is the master regulator of the feed forward loop leading to the expression of the flagella class 2 genes (fliL, flgA, flhB...). We measure here their activity with envZ expressed 0.2% arabinose or repressed 1% glucose.
