Design Notes

This sequence was optimized for expression in E. coli K12. As opposed to the eukaryotic Kozak seuquence on BBa_782004 TALE A from which the TALE in this sequence was inspired, the iGEM Calgary 2013 team placed a strong prokaryotic (B0034) RBS to express it. A GGGS*4 linker was placed between the two protein domains to prevent steric hindrance between the two functional domains. The KasI restriction site on the C terminus of the TALE and the N terminus of beta-lactamase is designed to allow user switch in new promoter-RBS-TALE sequences to improve modularity of our DNA detection system. Depending on the TALE sequence selected, on can detect any sort of DNA sequence of interest.
